Cornelia was born at Monticello in 1799. She was the fifth child of Martha Jefferson Randolph and Thomas Mann Randolph. Cornelia spent much of her early years at Monticello with her grandfather, Thomas Jefferson.

Cornelia learned about architecture from her grandfather. She practiced by sketching plans for the University of Virginia.

Cornelia never married. As an adult she lived with her brother, Thomas Jefferson Randolph, at Edgehill, his plantation located across the river from Monticello. Cornelia and her sister, Mary, established a school at Edgehill. Cornelia taught drawing, painting and sculpture. Cornelia died in 1871 and was buried at Monticello.
